Abstract
The difference of environmental pH requirements of Monascus ruber M7 at two different stages of growth and reproduction was studied, and pH was subsection controlled during spore culture, in order to reduce the influence of sporulation conditions on mycelium growth and increase spore yield. The results showed that the spores of strain M7 could germinate 100% when cultured for 8 h under pH 3.0-6.0, among them, pH 4.0-5.0 was the optimal and the germination was the fastest, spore germination was severely inhibited and the germination rate was very low under pH 7.0-8.0. Colony growth was faster at pH 4.0-6.0, among them, pH 5.0 was the optimal, and the colony diameter was the largest. However, the colony growth slowed down significantly at pH 3.0 and 7.0, and severely inhibited at pH 8.0, forming a small colony. The alkaline condition with pH>8.0 were extreme pH conditions, and spore germination and colony growth were completely inhibited. In the pH range of strain M7 growth, the sporulation capacity increased with the increase of pH, and the suitable sporulation pH range was 7.0-8.0. Based on the differences of pH requirement for growth and reproduction of strain M7, the spore production in PDB and CYB medium increased by 12-16 times and 3-4 times respectively by controlling the pH in the early stage of culture to be 5.0 and the pH in the later stage to be 7.0-8.0.
