A strain, L44, which could improve the output of conjugate lineleic acid (CLA), was isolated from kraut. The strain was identified belongs to Lactobacillus acidophilus by a series of morphological and biochemical characteristics analysis and 16S rDNA, which was named as L44. Cultural conditions in a bid for higher production of conjugate lineleic acid of L44 were optimized by five factors single-factor design and quadratic regression orthogonal rotary test. The determined conditions were incubation for 24h at 37℃ with pH value 7, and inoculum size 10% and the filling rate of liquid was 150ml. The production of CLA increased up to 107.0989μg/ml from 95.1425μg/ml.
