The intracellular polysaccharides of Armillariella tabescens by submerged culture were extracted by application of combined neutral protease and cellulase.The results showed that the extraction yield could be increased by application of complex enzymes.By single-factor and orthogonal tests,the optimum polysaccharides extraction conditions were obtained as follows: ratio of material and water 1∶40,and the first step treated with cellulose,the amount of cellulase 2%,pH value 5.5,the extraction temperature 45℃,the extracting time 1h,followed with addition of neutral protease 1.2%,pH value 7.2,the extracting temperature 55℃ and the extracting time was 2.5h.The UV spectrophotometry revealed that the intracellular polysaccharides of Armillariella tabescens would be no protein absorption after three times purification.
