摘要

芽孢杆菌在以褐藻酸钠为唯一碳源的培养基中进行驯化培养后,可在褐藻酸钠的诱导下分泌胞外褐藻胶裂解酶,经过发酵实验,其最佳氮源为蛋白胨和尿素,褐藻酸钠的较适浓度为0.5%~0.6%,在培养初始pH值为7.5、32℃、180r/min的条件下摇床发酵36h~84h时,产酶效果较好。

Abstract

A Bacillus sp.Stain was cultured in the solid medium with sodium alginate as the sole carbon source.The colony with transparent circle which indicated the production of alginate lyase was chosen as the initial strain and used to ferment the medium with sodium alginate.It was found that the optimal conditions of producing alginate lyase were as follows.The nitrogen sources are peptone and urea.The optimal content of sodium al-ginate is 0.5 %~0.6%.The alginate lyase can be well produced with initial pH 7.5 of liquid medium under the temperature 32℃ and rotation rate 180 r/min after fermentation for 36h~84h.
